LENS™ Technology for Fragment-based Lead Discovery

ActiveSight's LENS Technology combines complementary FBLD methods along with iterative Lead Optimization to quickly generate tightly binding Lead Compounds.

LENS, or Lead Evolution via Novel Synergies, is ActiveSight's proprietary Fragment-based Lead Discovery (FBLD) platform. LENS begins with ActiveSight's libraries of weakly binding Fragments (left) which bind with low affinity to the target protein of interest. To detect binding, ActiveSight employs complementary fragment screening technologies such as Structural Biology and Advanced Binding Detection Methods such as Surface Plasmon Resonance and Calorimetry. The use of multiple technologies ensures that a thorough screening of the fragment library has been performed, and that false positives and negatives are detected. With the help of Automated Software developed in-house structures of the fragment hits are analyzed and the information results in the design of libraries which are then synthesized utilizing state-of-the-art Parallel Chemistry. Through feedback-based iterations of these steps, tightly binding compounds are generated, which then undergo Cutting Edge Diagnostics to optimize in vitro and in vivo parameters. This multi-parametric and feedback-based approach results in a tightly binding Lead Compound (right) that is more likely to succeed in the clinic.
